The rumen is a complex microbial system of substantial importance in terms of greenhouse gas emissions and feed efficiency. This study proposes combining metagenomic and host genomic data for selective breeding of the cow hologenome toward reduced methane emissions. We analyzed nanopore long reads from the rumen metagenome of 437 Holstein cows from 14 commercial herds in 4 northern regions in Spain. After filtering, data were treated as compositional. The large complexity of the rumen microbiota was aggregated, through principal component analysis (PCA), into few principal components (PC) that were used as proxies of the core metagenome. The PCA allowed us to condense the huge and fuzzy taxonomical and functional information from the metagenome into a few PC. Bivariate animal models were applied using these PC and methane production as phenotypes. The variability condensed in these PC is controlled by the cow genome, with heritability estimates for the first PC of ~0.30 at all taxonomic levels, with a large probability (>83%) of the posterior distribution being >0.20 and with the 95% highest posterior density interval (95%HPD) not containing zero. Most genetic correlation estimates between PC1 and methane were large (≥0.70), with most of the posterior distribution (>82%) being >0.50 and with its 95%HPD not containing zero. Enteric methane production was positively associated with relative abundance of eukaryotes (protozoa and fungi) through the first component of the PCA at phylum, class, order, family, and genus. Nanopore long reads allowed the characterization of the core rumen metagenome using whole-metagenome sequencing, and the purposed aggregated variables could be used in animal breeding programs to reduce methane emissions in future generations.  相似文献

ABSTRACT

Concern over increasing water scarcity has led to the introduction of the concept of agricultural water productivity and an emphasis on interventions to achieve ‘more crop per drop’. Yet, a strong debate continues on how the concept is to be defined and used. Drawing largely from the irrigation literature, the origins of the concept and its methodological developments are reviewed, and its use in applied work over two decades is discussed. Based on this analysis of conceptual and applied research, key insights into the concept’s contributions and limitations are presented, as well as opportunities for further refinements.  相似文献

Thermodynamic measurements on BaMoO4, BaMoO3 and BaMo3O10 are reported, that served as input for the development of a thermodynamic model of the Ba-Mo-O system using the CALPHAD methodology. The valence states of molybdenum in BaMoO4 and BaMoO3 were confirmed to be VI and IV, respectively, from X-ray Absorption Near Edge Structure Spectroscopy measurements at the Mo K-edge. The heat capacity at low temperatures of these compounds was obtained from thermal-relaxation calorimetry. Phase equilibrium data in the BaMoO4-MoO3 section were also measured, and the transition enthalpy associated with the peritectic decomposition of BaMo3O10 was determined using Differential Scanning Calorimetry. The developed thermodynamic model used the compound energy formalism for intermediate compounds, and an ionic two-sublattice model for the liquid phase. The optimized Gibbs energies were assessed with respect to the known thermodynamic and phase equilibrium data. A good agreement is generally obtained, but a number of ill-defined data were also identified.  相似文献

A set of novel hydrazone derivatives were synthesized and analyzed for their biological activities. The compounds were tested for their inhibitory effect on the phosphorylating activity of the protein kinase CK2, and their antioxidant activity was also determined in three commonly used assays. The hydrazones were evaluated for their radical scavenging against the DPPH, ABTS and peroxyl radicals. Several compounds have been identified as good antioxidants as well as potent protein kinase CK2 inhibitors. Most hydrazones containing a 4-N(CH3)2 residue or perfluorinated phenyl rings showed high activity in the radical-scavenging assays and possess nanomolar IC50 values in the kinase assays.  相似文献

There is currently considerable interest in developing stiff, strong, tough, and heat resistant poly(lactide) (PLA) based materials with improved melt elasticity in response to the increasing demand for sustainable plastics. However, simultaneous optimization of stiffness, strength, and toughness is a challenge for any material, and commercial PLA is well-known to be inherently brittle and temperature-sensitive and to show poor melt elasticity. In this study, we report that high-shear mixing with cellulose nanocrystals (CNC) leads to significant improvements in the toughness, heat resistance, and melt elasticity of PLA while further enhancing its already outstanding room temperature stiffness and strength. This is evidenced by (i) one-fold increase in the elastic modulus (6.48 GPa), (ii) 43% increase in the tensile strength (87.1 MPa), (iii) one-fold increase in the strain at break (∼6%), (iv) two-fold increase in the impact strength (44.2 kJ/m2), (v) 113-fold increase in the storage modulus at 90°C (787.8 MPa), and (vi) 103-fold increase in the melt elasticity at 190°C and 1 rad/s (∼105 Pa) via the addition of 30 wt% CNC. It is hence possible to produce industrially viable, stiff, strong, tough, and heat resistant green materials with improved melt elasticity through high-shear mixing.  相似文献

In the context of the high-level radioactive waste disposal CIGEO, the corrosion rate due to microbially influenced corrosion (MIC) has to be evaluated. In France, it is envisaged to dispose of high- and intermediate-level long-lived radioactive waste at a depth of 500 m in a deep geological disposal, drilled in the Callovo-Oxfordian claystone (Cox) formation. To do so, a carbon steel casing will be inserted inside disposal cells, which are horizontal tunnels drilled in the Cox. A specific cement grout will be injected between the carbon steel casing and the claystone. A study was conducted to evaluate the possibility of MIC on carbon steel in the foreseeable high radioactive waste disposal. The corrosiveness of various environments was investigated at 50°C and 80°C with or without microorganisms enriched from samples of Andra's underground research laboratory. The monitoring of corrosion during the experiments was ensured using gravimetric method and real-time corrosion monitoring using sensors based on the measurements of the electrical resistance. The corrosion data were completed with microbiological analyses including cultural and molecular characterizations.  相似文献

Electron emission characteristic, electrical conductivity of polycrystalline mayenite (12CaO·7Al2O3) electride, formation of [Ca24Al28O64]4+(e)4 framework as a function of phase content, and microstructure have been investigated. The mayenite microstructure was investigated using high-resolution transmission microscopy which revealed the type cage structure of 12CaO·7Al2O3 partially filled by extra-framework oxygen ions. Incorporation of electrons by means of carbon ion template 12CaO·7Al2O3 produces complex structure, and an incomplete ion template 12CaO·7Al2O3 structure consisting of mixture of a [Ca24Al28O64]4+(e)4 and [Ca24Al28O64]4+(O2−)2 framework had a direct effect on the electron emission. Surface chemistry and stability of the 12CaO·7Al2O3 electride have been studied using x-ray photoelectron spectroscopy. The work function of phase pure 12CaO·7Al2O3 electride was determined from direct thermionic emission data and compared to the measurement from ultraviolet photoelectron spectroscopy (UPS). Depending on the extent of ion template of 12CaO·7Al2O3 structure, a work function of 0.9–1.2 eV and 2.1–2.4 eV has been measured and thermionic emission initiating at 600°C.  相似文献
